摘要
目的通过体外实验,研究芦荟大黄素(aloe-emdion,AE)是否能阻断肺炎链球菌(Streptococcuspneumoniae,S.pn)触发人肺Ⅱ型上皮细胞A549微丝肌动蛋白(filamentousactin,F-actin)细胞骨架重排。方法采用F-actin特异性荧光染料观察S.pn作用A549细胞前后的F-actin细胞骨架重排情况并用AE预处理A549细胞,观察其与F-actin细胞骨架重排关系。结果用AE预处理A549后S.pn侵袭数为(22±4)CFU/孔,而没有用AE预处理的A549S.pn侵袭数为(138±21)CFU/孔,经两样本均数t检验,有显著差异(P<0.01)。结论AE对S.pn侵袭人肺Ⅱ型上皮细胞A549F-actin细胞骨架的重排有抑制作用。
Objective To study the effect of aloe-emdion on blocking the F-actin rearrangements in the Streptococcus pneumoniae invaded A549 cells. Metho d F-actin rearrangements was observed by fluorescent staining method. Results The invasion rates of S.pneumoniae to A549 cells pretreated with aloe-emodin and without treatment were (22±4)CFU/well and (138±21)CFU/well respectively(P<0.0 1). Intact S.pneumoniae promoted F-actin rearrangements. Aloe-emodin was able to prevent S.pneumoniae from invasion into A549 cell. Conclusion Aloe-emodin can inhibited the typeⅡ pneumocytes filamentous actin rearrangements induced by St reptococcus pneumoniae.
