摘要
目的探讨硫代磷酸反义寡脱氧核苷酸对乙型肝炎病毒细胞的影响。方法设计合成HBV5个硫代反义寡聚核苷酸(S-asODN)靶序列。应用ELISA方法、MTT比色法、观察S-asODN对HBsAg、HBeAg抗原表达及HepG22215细胞毒性的影响。结果不同靶位点的S-asODN对HBsAg、HBeAg表达都有显著的抑制作用,在浓度为20μmol/L时,对细胞无明显杀伤作用。结论S-asODN对HepG22215的HBsAg、HBeAg的抑制作用为抗HBV治疗提供了新的途径。
Objective To explore the effects of antisense phosphorothioat e oligodeoxynucleotides on HBV infected hepatic cells. Methods According to gen e structre of HBV,5 antisense phosphorothioats oligodeoxy- nucleotides (S-as ODN ) were designed and synthesized. The expression of HBsAg and HBeAg levels were d etermined by ELISA. Cytotoxicity was measured by MTT colorimetrie assays. Resul ts The 5 S-as ODN could specificly inhibit the production of HBsAg and HBeAg in HepG22215 cells. Under 20 μmol/ L, the cytotoxicity effect of S-as ODN to HepG 22215 cells was below 10%. Conclusion These results suggest that S-as ODN may b e developed as the new therapy for anti-HBV.
