MTX致小鼠组织DNA损伤的研究

摘要 目的为进一步了解甲氨蝶呤(methotrexate,MTX)的作用机制,探测一次给药后不同时间点小鼠组织DNA损伤的修复情况。方法用单细胞凝胶电泳技术检测MTX腹腔注射染毒1、3、6、12、24h后对小鼠肝、脾、骨髓、胸腺、肾、睾丸、胃和外周血淋巴细胞的DNA损伤作用。结果腹腔注射5mg/kgMTX可诱发小鼠脾细胞、骨髓细胞、胸腺细胞和外周血淋巴细胞的DNA单链断裂。结论MTX可致小鼠体内多脏器细胞的DNA单链断裂,不同细胞对MTX的易感性不同。 Objective To further understand the mechanism of MTX and to i nvestigate DNA damage induced by MTX. Methods Liver, spleen, bone marrow, thymu s, kidney, testicle, stomach and peripheral lymphocytes samples of mice were pre pared at 1,3,6,12 h, and 24 h after MTX intraperitoneal injection of MTX. DNA da mage was monitored by using the alkaline single cell gel electrophoresis (comet assay). Results Significant increase in DNA migration and comet frequency were observed in the spleen, thymus, bone marrow and peripheral lymphocytes samples o f the mice treated with MTX at a dose of 5 mg/kg. Conclusion DNA SSBs could be induced by MTX in some organs of mice. The degrees of DNA damage in different organs were varied.
