鼻腔非霍奇金淋巴瘤EB病毒感染与Survivin基因表达的关系

Association between Epstin-Barr virus infection and Survivin gene expression in nasal non-Hodgkin's lymphoma

摘要 目的观察EB病毒(Epstin Barrvirus,EBV)RNA、膜蛋白(LMP1)和凋亡抑制基因Survivin在鼻腔非霍奇金淋巴瘤(non Hodgkin’slymphoma,NHL)中的表达,探讨其相互间的关系及其在鼻腔NHL发生发展中的作用。方法用原位分子杂交技术检测29例鼻腔NHL中EBV编码的RNA(EBER1/2),用免疫组织化学技术(SP法)检测LMP1、Survivin蛋白表达。结果23例(79.31%)EBER1/2阳性,14例(48.28%)LMP1阳性;19例(65.52%)Survivin阳性,Survivin阳性表达在中——高度恶性组(18/23例)与低度恶性组(1/6例)中有显著性差异(P<0.05);EBV阳性组与EBV阴性组之间Survivin表达差异无显著性(P>0.05)。结论鼻腔NHL中有较高比例的EBV感染,Survivin表达阳性率与鼻腔NHL的恶性程度有关,Survivin基因可能通过抑制肿瘤细胞凋亡而参与NHL的发生发展,但未发现EBV感染与Survivin基因表达之间有明显相关性。 Objective To observe the presence of Epstin-Barr virus (EBV) and expression of Survivin gene in nasal non-Hodgkin's lymphoma(NHL). Methods In situ hybridization (ISH) was used to detect EBV-encoded RNA (EBER1/2) and immunohistochemistry staining was used to examine the expression of EBV-latent membrane protein (LMP-1) and Survivin gene in the 29 cases. Results Positive expression rates of EBER 1/2 and LMP-1 were 79.31% and 48.28%, respectively. Survivin expression was positive in 19 nasal NHL (65.52%) while no expression of Survivin was detected in the 10 reactive lymphoid tissues. In the low grade group, Survivin positive expression rates was 16.67%. Whereas in high grade group, Survivin positive expression rates was 78.26%. Both differences were significant (P<0.05). Survivin expression was not significantly different between EBER1/2 positive and negative cases, nor was it significantly different between LMP-1 positive and negative cases (P>0.05). Conclusion A higher percentage of EBV infection was detected in the nasal NHL. Survivin expression were associated with the degree of malignancy.Survivin gene may play a role in the pathogenesis of the nasal NHL by affecting cell apoptosis. EBV infection was not significantly associated with Survivin gene expression in the nasal NHL.
