摘要
采用RAPD技术,用524个随机引物对3个抗病品种和4个感病品种的基因组DNA进行扩增,发现3个引物能在抗、感品种间检测到稳定的多态性。这3个引物及它们产生的多态片段是S3471220bp、S372875bp和S3751360bp,其中S3471220bp和S372875bp在感病品种上呈显性扩增,在抗病品种上无扩增,相反S3751360bp在感病品种上无扩增,在抗病品种上呈显性扩增。初步判断这3个特异带与小麦赤霉病抗性有关。
524 random primers were used in RAPD analysis to amplify genomes of three scab-resistant varieties and four scab-susceptible varieties and three of them were found to show stable polymorphism in both the scab-resistant and -susceptible varieties.The polymorphic bands amplified with the three primers were S347_(1 220 bp),S372_(875 bp) and S375_(1 360 bp).Of the bands,S347_(1 220 bp) and S372_(875 bp) were found in all the susceptible varieties but not in all the resistant varieties whereas S375_(1 360 bp) was not found in all the susceptible varieties but found in all the resistant varieties. It was preliminarily concluded that these three specific bands related to scab resistance.
