摘要
目的研究神经再生抑制性蛋白Nogo在不同发育阶段大鼠中枢神经系统(CNS)中的表达。方法采用RT-PCR方法半定量分析不同发育阶段大鼠(胚胎期14d ̄成年,共7个时间点)脊髓中Nogo mRNA的表达变化。结果Nogo-A mRNA和Nogo-B mRNA在胚胎期14d就可以检测到,在所检查的时间点内,Nogo-A mRNA和Nogo-B mRNA的扩增表现出很恒定的水平,其表达并未有任何特定的时间模式;Nogo-C mRNA的表达比前两者稍晚,在胚胎期21d检测到。结论Nogo在神经系统发育期就已存在,说明Nogo-A分子与髓鞘的成熟化进程关系可能并不很密切,提示Nogo分子在发育阶段具有不同于神经再生抑制作用的其他功能。
Objective To study the expression of Nogo mRNA in the rat's central nervous system(CNS) at various developing stages. Methods Sem i-quantitative RT-PCR was used to study the expression and level of Nogo mRNA in the rat spinal cord of different developing stages (from embryonic day 14 to ad ult, 7 stages in total). Results Nogo-A mRNA and Nogo-B mRNA were positive from embryonic day 14 and remained stable during all the check points without t ime-dependent manner. Nogo-C mRNA was found after embryonic day 21 later than th e Nogo-A and Nogo-B mRNA. Conclusion Nogo is present in the developing CNS and has no close relation with myelin maturation, which indicates that Nogo mayb e have more function than the neurite growth inhibition, especially during the d eveloping CNS.
