摘要
目的观察阿司匹林(ASA)对体外培养的大鼠血管平滑肌细胞增殖和磷酸化p44/42丝裂素活化蛋白激酶(MAPK)表达的影响。方法体外培养SD大鼠血管平滑肌细胞,应用不同浓度的ASA(1mmol/L、2mmol/L、5mmol/L及10mmol/L)分别作用并设立对照进行比较,采用MTS/PES法确定血管平滑肌细胞的增殖状态,利用p44/42磷酸化抗MAPK抗体的蛋白免疫印迹法测定磷酸化p44/42MAPK蛋白表达。结果各组的细胞增殖率分别为对照组0.79±0.14;ASA1mmol/L组0.81±0.16;ASA2mmol/L组0.83±0.08;ASA5mmol/L组0.60±0.07;ASA10mmol/L组0.35±0.05。统计分析显示5mmol/L和10mmol/L的ASA分别能明显抑制大鼠血管平滑肌细胞的增殖(P<0.05)。ASA对大鼠血管平滑肌细胞的磷酸化p44/p42MAPK蛋白表达有显著的抑制作用(P<0.05)。结论ASA对体外培养的大鼠血管平滑肌细胞增殖和磷酸化p44/42MAPK蛋白表达均有明显的抑制作用。
Objective To observe the effects of aspirin on the proliferat ion in vitro of rat vascular smooth muscle cells (VSMCs) and the activity of pho sphorylation p44/p42 mitogen-activated protein kinase (MAPK). Methods Rat aortic VSMCs were cultured in vitro and treated with aspirin at the concentratio ns of 1 mmol/L, 2 mmol/L, 5 mmol/L and 10 mmol/L and compared with controls. The ratio of cell proliferation was determined by non-radioactive MTS/PES assay. Th e expression of p44/42 MAPK phosphorylation protein was evaluated by the immunob loting technique using anti-p44/42 phospho-MAPK antibody. Results The rati o of VSMCs proliferation was 0.79±0.14 in the control group, and 0.81±0.16, 0. 83±0.08, 0.60±0.07, 0.35±0.05 in aspirin groups corresponding to aspirin conc entrations of 1mmol/L, 2 mmol/L, 5 mmol/L and 10 mmol/L, respectively. When the concentrations exceeded 5 mmol/L, aspirin significantly inhibited the VSMC proli feration (P<0.05). The expression of p44/42 MAPK phosphorylation protein was sig nificantly inhibited by aspirin. Conclusion Aspirin can inhibit VSMCs proli feration. Arrest of VSMCs cell proliferation may be an important mechanism of th e beneficial effect of aspirin in acute coronary syndromes.
