摘要
目的:探讨胃癌中CyclinG1、CyclinD、p21、p16、p53、bcl-2等基因的表达水平、细胞周期变化和凋亡情况。方法:建立胃癌模型;免疫组化法检测小鼠胃癌中CyclinG1、CyclinD、p21、p16、p53、bcl-2等基因蛋白表达,RT-PCR技术检测CyclinG1mRNA的表达;流式细胞术检测小鼠胃癌细胞的凋亡率和细胞周期。结果:阳性组的CyclinG1、CyclinD表达明显高于正常组和化疗组(P<0.05),p16则相反;各组p21、bcl-2的表达无明显差异;阳性组p53的表达为阴性,而化疗组与正常组表达无明显差异(P>0.05);阳性组中CyclinG1mRNA的表达水平明显高于正常组和化疗组(P<0.05);化疗组和阳性组的细胞凋亡率均较正常组增高(P<0.05),且化疗组的凋亡率比阳性组明显增高(P<0.01)。结论:CyclinG1、CyclinD高表达、p16低表达可能与胃癌的发生、发展有关。
Objective: To study expression of Cyclin G1?Cyclin D?p21?p16?p53?bcl-2,apoptosis and change of cell cycle in mice-borne gastric carcinoma. Methods: Mice models with gastric carcinoma cell were established. The protein expression of Cyclin G1, Cyclin D, p21, p16, p53, bcl-2 was measured using immunohistochemical method; the expression of Cyclin G1 mRNA was measured with RT-PCR; Rate of apoptotic cells and cell cycle were detected by Flow cytometry. Results: Expression of Cyclin G1 and Cyclin D in positive groups was significantly higher than in normal and chemotherapy groups(P<0.05), but that of p16 was reverse. Expression of p21 and bcl-2 showed no difference (P>0.05) in all groups; Expression of p53 in positive groups was negative, in other two groups it showed no difference (P<0.05). The expression of bcl-2 showed no difference(P>0.05); Rate of apoptotic cell in chemotherapy group and postive group was higher than that in normal group (P<0.05), and chemotherapy group was obviously higher than normal group(P<0.01). Conclusions: Gastric carcinoma relates to a higher expression of Cyclin G1, Cyclin D and lower expression of p16.
