摘要
以子实体及菌丝体为试材对香菇、平菇及金针菇进行多糖提取技术研究。子实体按照粗品制备、半精品纯化、胃蛋白酶解3个步骤提取多糖。100g香菇、平菇及金针菇子实体中纯多糖平均提取量分别为15.0、7.8和26.6mg,并用咔唑反应、纸层析法对其进行鉴定。50g香菇、金针菇菌丝中多糖的平均提取量分别为15.2和20.4mg。对2个品种多糖提取率进行比较,结果表明:菌种液体培养提取多糖产量大且以金针菇最佳。采用子实体及液体培养菌丝提取多糖方法是有效的,且菌丝液体培养提取多糖产量高于子实体提取多糖产量。
In this paper, the extraction technology of polysaccharide from Lentinus edodes, Pleurotus ostreatus and Flammulina velueipes by means of fruit body extraction and cultivated mycelium on liquid medium was studied.The extraction procedures of polysaccharide of fruit body was as follow:preparation of crude polysaccharide, purification of semi-purified polysaccharide, pepsin hydrolyze etc. The average content of pure polysaccharide in fruit body from Lentinus edodes,Pleurotus ostreatus and Flammulina velueipes was 15?mg/100?g,7.8?mg/100?g and 26.64?mg/100?g,respectively.The polysaccharide was identified by means of carbazole reaction and filter paper chromatography. The average yield of polysaccharide in cultivated mycelium on liquid medium of Lentinus edodes and Flammulina velueipes was 15.2?mg/50?g and 20.4?mg/50?g.And compared the polysaccharide yield of Lentinus edodes and Flammulina velueipes, the results showed: the polysaccharide yield of Flammulina velueipes extracted from cultivated mycelium on liquid medium was the greatest. The extraction technology of polysaccharide by means of fruit body extraction and cultivated mycelium in liquid medium was confirmed, and the polysaccharide yield extracted from mycelium on liquid medium was higher than extracted from fruit body.
