摘要
目的建立用过硫酸铵消化尿样30℃控温反应测定尿碘的砷铈催化分光光度法。方法对本方法不同反应时间标准曲线各点的吸光度值及反应30min时方法的灵敏度、精密度、准确度进行研究,并比较了控温(30min反应时间)和室温两种反应条件测定的33份人尿碘浓度。结果本方法标准曲线剂量范围为0~300μg/L,反应30min时吸光度值范围为0.166~1.064;检验下限为5.5μg/L;测定低、高两种碘浓度的国家冻干人尿标准物,其变异系数分别为2.0%和3.5%,其相对偏差分别为6.2%和-2.2%;控温和室温两种反应条件测定的尿碘浓度具有高度相关性和良好的一致性。结论本方法是一种灵敏、准确、操作简便测定尿碘的好方法。
Objective To establish a method for measuring iodine in urine by As^(3+)-Ce^(4+) catalytic spectrophotometry using ammonium persulfate digestion in controlled temperature condition.Methods Optical density of calibration curve in different reaction time and methodological assessment(30 min reaction time)were observed.The comparisons of iodine concentration were also studied in 33 samples tested by two different reaction temperature conditions(30 ℃ and room temperature).Results The detection range of calibration curve and its optical density(30 min reaction time)were 0~300 μg/L and 0.166~1.064,respectively.The lowest limit of detection was 5.5 μg/L.The coefficients of variation were 2.0% and 3.5%,the relevant deviation was 6.2% and -2.2% when measuring'standard materials'containing low or high iodine concentration.Significant correlation and consistency between iodine concentrations measured by two different reaction conditions were obtained.Conclusion This method is sensitive,accurate and easily performed for measuring urinary iodione.
