FasR和FasL基因在口腔癌组织中表达与凋亡关系研究

摘要 目的 探讨口腔癌组织及正常组织的凋亡与FasR、FasL基因表达的关系。方法 利用免疫组织化学法检测口腔癌组织及癌旁正常组织中FasR、FasL基因的表达,采用原位末端标记(TUNEL)法检测口腔癌组织标本中的凋亡情况。结果 FasR基因在口腔癌组织和正常口腔组织中的阳性表达率分别为5 6.7% (3 8/67)和83 .6% (3 8/4 2 ) ,FasR基因在口腔癌组织中的阳性表达率显著低于正常口腔组织(P <0 .0 0 1)。FasL基因在口腔癌组织和正常口腔组织中的阳性表达率分别为85 .7% (5 6/67)和16.7% (7/4 2 ) ,口腔癌组织中FasL基因的表达显著高于正常口腔组织(P <0 .0 0 1)。口腔癌组织细胞凋亡指数显著低于正常口腔组织(P <0 .0 0 1)。FasR阳性的口腔癌组织细胞凋亡指数显著高于FasR阴性者(P<0 .0 0 1) ,而FasL阳性的口腔癌组织细胞凋亡指数显著低于FasL阴性者(P <0 .0 5 )。结论 FasR和FasL基因在口腔癌组织细胞凋亡过程中存在着协同或拮抗作用的关系。 Objective To investigate the inter-relationship between the exp ressions of FasR and FasL genes and spontaneous apoptosis in oral carcinoma. Methods Oral carcinoma and oral tissue were examined for the e xpressions of FasR and FasL genes by immunohitochemistry, apoptosis by in-situ TUNEL. Res ults The detection rates of FasR in oral carcinoma and oral tissue wer e 56.7%(38/67) and 83.6%(38/42) respectively, expression of FasR in oral carci noma was significantly lower than that in oral tissue(P<0.001). The delectio n rates of FasL in oral carcinoma and oral tissue were 85.7%(56/67) and 16.7%(7 /42) reapectively, expression of FasL in oral carcinoma was significantly highe r than that in oral tissue(P<0.001). The apoptotic index of cells in oral ca rcinoma was significantly lower than that in oral tissue(P<0.001). The apopt otic index of cells of FasR positive oral carcinoma was significantly higher tha n that of FasR negative(P<0.001). The apoptotic index of cells of FasL poiti ve oral carci noma was significantly lower than that of FasL negative(P<0.05). C onclusion Expressions of FasR and FasL gene has synergistic or antagonistic action in apoptosis of oral carcinoma.
