胡黄连提取物对大鼠脑缺血再灌注后半暗带区细胞凋亡及bcl-2表达的影响 被引量:16

摘要 目的:研究胡黄连提取物对脑缺血再灌注半暗带神经细胞凋亡及其相关基因表达的影响。方法:Wister大鼠随机分为假手术组、模型组、尼莫地平组和治疗组。利用流式细胞仪分别观察缺血1.5h再灌注用药5d后半暗带神经细胞凋亡及相关基因bcl-2蛋白表达。结果:与模型组比较,胡黄连组缺血半暗带凋亡细胞百分数明显降低(P<0.05),bcl-2蛋白表达显著升高(P<0.05)。结论:胡黄连提取物可能通过促进bcl-2基因的表达,对脑缺血再灌注模型半暗带的细胞凋亡产生抑制作用。 Objective:to investigate the effects of extraction huhuanglian on apoptosis and the expression of relating gene bcl-2 in penumbra area of local cerebral ischemic reperfusive model rats.Methods: Wister rats were randomly divided into sham operated group, model group, nimodipine group and treated group. The middle cerebral arteries of rats were occluded for 1.5 hours. After 5 days of treatment, apoptosis and bcl-2 expression were measured by flow cytometer. Result: compared with model group,extraction huhuanglian treated group showed a distinct decrease (P<0.05) in apoptosis and increase in bcl-2 expression (P<0.05). Conclusion:extraction huhuanglian can prevent apoptosis in penumbra area of local cerebral ischemic reperfusive model rats by enhancing bcl-2 expression.
