三氧化二砷对人结肠癌裸鼠移植瘤端粒酶活性及其催化亚单位表达的影响 被引量:11

摘要 目的:研究三氧化二砷(As2 O3 )注射液对人结肠癌裸鼠移植瘤端粒酶活性及其催化亚单位人端粒逆转录酶(hTERT)表达的影响。方法:建立人结肠癌裸鼠移植瘤模型,随机分为生理盐水组、As2 O3 低剂量(2 .5mg·kg-1)组、As2 O3 高剂量(5mg·kg-1)组及5 - FU(2 0mg·kg-1)组。腹腔注射As2 O3 ,利用银染 端粒重复扩增(TRAP)法测定人结肠癌裸鼠移植瘤端粒酶活性、RT PCR法检测hTERT的表达。结果:生理盐水组及5 -FU组端粒酶活性及其催化亚单位表达不受影响,不同剂量As2 O3 均明显抑制端粒酶活性及其催化亚单位表达。结论:As2 O3 注射液对人结肠癌裸鼠移植瘤端粒酶活性及其催化亚单位的表达有抑制作用。 Objective To study the influence of As_2O_3 on human telomerase reverse transcriptases(hTERT) expression and tolemerase activity of human colonic carcinoma xenograft in nude mice. Methods To establish human colonic carcinoma xenograft model, As_2O_3 was intravenousely injected into nude mice.Tolemerase activity was detected by silver stain with TRAP. The expression of hTERT was detected by reverse transcription polymerase chain reaction (RTPCR). Result The tolemerase activity and hTERT expression of NS control group and 5FU group were highly expressed while those of low and high As_2O_3 concentration groups were both obviously inhibited. Conclusion As_2O_3 can greatly inhibit tolemerase activity and hTERT expression of human colonic carcinoma xenograft in nude mice.
