bFGF对大鼠牵张性脊髓损伤后脊髓神经元影响的实验研究 被引量:1

摘要 目的探讨碱性成纤维细胞生长因子(bFGF)对牵张性脊髓损伤后神经元影响的作用。方法大鼠脊髓T13~L2经牵张损伤,CSEP监测P1~N1波幅下降至术前波幅70%后,于损伤平面以下经蛛网膜下腔置细导管,治疗组分别于术后即刻、1、2、3、4、8、12及24h经细导管注入bFGF溶液20μl(含bFGF20μg),对照组在相同时间注入等量生理盐水,然后于术后1d、4d、7d、14d及21d处死取材(n=4)。应用行为学及CSEP检查大鼠功能恢复情况,应用尼氏染色法观察神经元及尼氏体密度,并用计算机图像分析系统进行定量分析。结果bFGF治疗组大鼠的神经功能恢复与对照组相比差异有显著意义。尼氏染色的图像分析显示:bFGF治疗组的神经元截面积及尼氏体密度与对照组比较差异有显著意义(P<0.01)。结论bFGF对大鼠牵张性脊髓损伤后脊髓功能及神经元形态的恢复有明显的促进作用。 Objective To explore the effect of bFGF on neurons after tractive spin al cord injury in rats. Methods With decreasing of the P1~N1 wave control 70% through CSEP monitor, The T13~L2 spinal cord of rats were injured by traction,a nd a thin plastic tube was situated in subarachnoid space below the injured leve l for perfusion. The bFGF-treated rats were received bFGF 20μg(20μl liquid)fro m the tube at once,1?2?3?4?8?12 and 24 hours after injury respectively. The sali ne-treated rats were received the equal volume of normal saline at the same time ,then rats were killed in1?4?7?14 and 21 days postoperatively,neurological funct ion change of rats was tested with combined behavioral score(CBS)and cortical so matosensory evoked potential(CSEP),section area and Nissl body density of neuron was observed with nissl stained and was analyzed quantitatively with image syst em of computer. Results Neurological function recovery,section area and Nissl b ody density of neuron had a marked difference in bFGF-treated group than in cont rol group (P<0.01). Conclusion bFGF has a significant improvement in recovery o f spinal cord function and neuron morphology.
