摘要 【目的】探讨Persephin对6鄄羟基多巴(6鄄OHDA)损伤神经干细胞的保护作用。【方法】用RT鄄PCR法从人胚胎脑获得PersephincDNA,构建重组Persephin腺病毒。用Westernblot方法分析Persephin基因的表达,Hoechst染色和流式细胞术测定6鄄羟基多巴对神经干细胞凋亡的诱导作用。【结果】从人胎脑成功克隆了人PersephincDNA,构建了其腺病毒载体。Westernblot证实重组Persephin腺病毒在神经干细胞中的表达,表达的Persephin可抑制6鄄羟基多巴诱导的神经干细胞凋亡。【结论】Persephin对6鄄羟基多巴损伤神经干细胞具有保护作用。 Objective] To observe the protective effect of Persephin on C17.2 neural stem cells damage induced by 6-hydroxydopamine (6-OHDA). [Methods] Human Persephin cDNA was amplified by RT-PCR from human embryo brain. Recombinant Persephin adenovirus was constructed. Recombinant Persephin adenovirus production was observed by Western blot analysis. The apoptosis of C17.2 neural stem cells induced by 6-hydroxydopamine was determined by flow cytometry and Hoechst stain. [Results] Human Persephin cDNA was successfully amplified by RT-PCR from human embryo brain. Recombinant Persephin adenovirus vector was successfully constructed. Persephin expression was detected by Western blot in C17.2 neural stem cells. The apoptosis rate of C17.2 neural stem cells induced by 6-hydroxydopamine was decreased by Persephin expression. [Conclusions]Persephin has the protective effect on C17.2 neural stem cells damage induced by 6-hydroxydopamine. This provides the basis for the further study of Persephin on function of neural stem cells and treating Parkinson's disease.
