低浓度二甲亚砜和第二信使调节剂混合物低温保存血小板的初步研究 被引量:3

摘要 目的:探讨低浓度DMSO(2%)和第二信使调节剂混合物(thrombosol)低温保存血小板的效果。方法:应用6%DMSO(对照组)、2% DMSO+thrombosol(实验组)为新鲜浓缩血小板的冰冻保护剂-80℃保存,1周后复温检测血小板计数、肾上腺素或凝血酶诱导的聚集反应、血小板表面CD41、CD42b、CD62p和CD35分子改变。结果:复温后,实验组血小板计数和凝血酶诱导的聚集反应明显高于对照组(P<0.05,P<0.01),但仍然低于新鲜血小板(P<0.01)。冰冻血小板CD62p表达较新鲜血小板显著升高(P<0.05,P<0.01),但实验组显著低于对照组(P<0.05)。与新鲜血小板相比,冰冻血小板肾上腺素诱导的聚集反应和CD42b表达均下降(P<0.05,P<0.01),但实验组与对照组间比较无明显差异。结论:与常规6%DMSO低温保存血小板相比,2% DMSO加第二信使调节剂混合物低温保存血小板的效果有明显改善。 Objective:To study the in vitro activity of platelets following cryopreservation with mixed solution of low concentration DMSO(2%) and second-messenger effectors. Methods: Fresh platelet concentrate was cryopreserved with 6% DMSO(control group) and mixed solution of 2% DMSO and second-messenger effector(treated group) for one week. The platelet count, adrenaline and thrombin induced aggregation, and expression level of CD41,CD42b ,CD62p and CD35 were measured in the thawed platelets.Results: Compared with the control group, treated platelets had higher platelet count (P< 0.05) and thrombin induced aggregation (P<0.01), though still lower than fresh platelets (P<0.01). Compared with fresh platelets, cryopreserved platelets showed a significant increase in CD62p expression (P<0.05,P<0.01), but the increase of treated platelets were significantly lower than that of control (P<0.05).Adrenaline induced aggregation and expression level of CD42b of cryopreserved platelets were lower than those of fresh platelets (P<0.05,P<0.01),and there were no significant difference between the control and treated platelets.Conclusion: The in vitro activity of platelets cryopreserved with mixed solution 2% DMSO and second-messenger effector is greatly improved compared with platelets cryopreserved with 6% DMSO.
