摘要
目的:探讨低浓度DMSO(2%)和第二信使调节剂混合物(thrombosol)低温保存血小板的效果。方法:应用6%DMSO(对照组)、2% DMSO+thrombosol(实验组)为新鲜浓缩血小板的冰冻保护剂-80℃保存,1周后复温检测血小板计数、肾上腺素或凝血酶诱导的聚集反应、血小板表面CD41、CD42b、CD62p和CD35分子改变。结果:复温后,实验组血小板计数和凝血酶诱导的聚集反应明显高于对照组(P<0.05,P<0.01),但仍然低于新鲜血小板(P<0.01)。冰冻血小板CD62p表达较新鲜血小板显著升高(P<0.05,P<0.01),但实验组显著低于对照组(P<0.05)。与新鲜血小板相比,冰冻血小板肾上腺素诱导的聚集反应和CD42b表达均下降(P<0.05,P<0.01),但实验组与对照组间比较无明显差异。结论:与常规6%DMSO低温保存血小板相比,2% DMSO加第二信使调节剂混合物低温保存血小板的效果有明显改善。
Objective:To study the in vitro activity of platelets following cryopreservation with mixed solution of low concentration DMSO(2%) and second-messenger effectors. Methods: Fresh platelet concentrate was cryopreserved with 6% DMSO(control group) and mixed solution of 2% DMSO and second-messenger effector(treated group) for one week. The platelet count, adrenaline and thrombin induced aggregation, and expression level of CD41,CD42b ,CD62p and CD35 were measured in the thawed platelets.Results: Compared with the control group, treated platelets had higher platelet count (P< 0.05) and thrombin induced aggregation (P<0.01), though still lower than fresh platelets (P<0.01). Compared with fresh platelets, cryopreserved platelets showed a significant increase in CD62p expression (P<0.05,P<0.01), but the increase of treated platelets were significantly lower than that of control (P<0.05).Adrenaline induced aggregation and expression level of CD42b of cryopreserved platelets were lower than those of fresh platelets (P<0.05,P<0.01),and there were no significant difference between the control and treated platelets.Conclusion: The in vitro activity of platelets cryopreserved with mixed solution 2% DMSO and second-messenger effector is greatly improved compared with platelets cryopreserved with 6% DMSO.
