块菌多糖对小鼠肿瘤及免疫系统的影响

Effects of Polysaccharide of Tuber sinica on Tumor and Immune System of Mice

块菌多糖（ｐｏｌｙｓａｃｃｈａｒｉｄｅｏｆＴｕｂｅｒｓｉｎｉｃａ，ＰＳＴ）是首次从中国块菌中提取的一种新的蛋白结合多糖，民间用于治疗胃癌。为了开发ＰＳＴ的药用价值，研究了其对小鼠肿瘤及免疫系统的影响，结果表明ＰＳＴ连续１０ｄｉｐ２５ｍｇ／ｋｇ，５０ｍｇ／ｋｇ能抑制小鼠Ｓ－１８０肉瘤以及ＥＡＣ肉瘤的生长，但不抑制体外培养Ｈｅｌａ细胞的增殖。ＰＳＴ明显增加小鼠脾脏重量，明显促进碳粒廓清速率，增加ＳＲＢＣ所致的小鼠迟发型足垫肿胀度，增加外周血中白细胞数及Ｔ淋巴细胞百分率，促进Ｔ淋巴细胞转化，提高小鼠血清中ＩｇＧ水平。

Polysaccharide of Tuber sinica (PST) is a new protein polysaccharide(MW ca.3×104) extracted from Tuber.sinica K.Tao et Liu,which was used in the treatment of stomach cancer in folk medicine.The present paper reported effects of PST on tumor and immune system of mice. The results showed that PST ip 12.5,25,50 mg/kg inhibited the sarcoma proliferation of S-180 and Ehrlich ascitic carcinoma(the inhibition percentage of 50 mg/kg dose group was 50.2%and 35.7%,respectively),but did not affect of proliferation of Hela cells in uitro,PST ip 12.5, 25,50 mg/kg increased weights of spleen of mice,enhanced the clearance rates of charcoal particles and delayed footpad swellings,PST also increased the number of leukocytes,T lymphocytes percentage and lymphocytes transformation of blood in mice,and promoted the level of immunoglobulin G.This study indicated that the tumorinhibiting activity of PST may be related to immunological function in host.
