Ox-LDL对内毒素血症小鼠肝、肺组织TNF-α含量的影响及其意义

摘要 目的研究氧化型低密度脂蛋白(OxLDL)注射对内毒素致炎效应的影响。方法OxLDL0.35mg/kg或1.4mg/kg静脉注射预处理小鼠30min后注射LPS1mg/kg,ELISA法测定肝、肺组织TNFα含量变化。结果与单纯注射LPS相比,OxLDL预处理小鼠肝、肺组织TNFα含量明显升高(P<0.01)。结论OxLDL能增强LPS的致炎作用,使小鼠肝、肺组织TNFα增加,其机制可能与清道夫受体(SR)封闭,LPS清除减少、过多的游离LPS大量激活炎症细胞有关。 Objective To study the effect of Ox-LDL injection on the inflammatory ability of LPS.Methods Mice were injected with 0.35mg/kg or 1.4mg/kg of Ox-LDL 30min before 1mg/kg of LPS injection, and TNF-αlevels in the liver and lungs of mice were measured with ELISA.Results TNF-αlevels in the liver and lungs of endotoxemia mice pretreated with Ox-LDL were higher than that of endotoxemia mice(P<0.01).Conclusion Ox-LDL could remarkably enhance the inflammatory effect of LPS, resulting augment of TNF-αlevels in the liver and lungs of endotoxemia mice. The mechanism may attribute to blockage of scavenger receptors by Ox-LDL, which resulted decrease of LPS elimination. As a result, more free LPS actived inflammatory cells.
