摘要
目的:探讨通腑抗炎汤对腹腔感染所致SIRS的作用机理。方法:36只大鼠随机分为模型组、治疗组及正常对照组。模型组、治疗组腹腔注射大肠杆菌(1ml/100g加佐剂10%BaSO41ml/100g)造成大鼠SIRS模型。观察大鼠15min~48h的行为活动、精神状态、食欲、呼吸、大小便及死亡率等一般情况,于造模后24h尾静脉取血测白细胞计数、IL-1、IL-6和IL-10的变化。48h后取动物肝、肾、肺、小肠等脏器标本进行光镜观察。结果:治疗组与模型组大鼠均出现一般情况恶化,白细胞计数、IL-1、IL-6、IL-10水平均高于正常对照组(P<0.05)。治疗组白细胞计数、IL-1、IL-6水平低于模型组,IL-10水平高于模型组。治疗组肝、肾、肺、小肠等脏器光镜下炎症性病理损害均明显轻于模型组。结论:通腑抗炎汤可抑制腹腔感染大鼠白细胞升高幅度,减轻肝、肾、肺、小肠等脏器的继发性炎症性病理损害。通过抑制促炎细胞因子、上调抗炎细胞因子的双向调节来减轻或阻断腹腔感染所致SIRS进程。
Objective:To evaluate the effect of Tongfu Kangyan decoction(TFKYD ) in treatment of SIRS rats induced by acute abdominal infection and explore the p ossible mechanism.Method:All the rats were divided randomly into model, treatmen t and normal groups.SIRS- MODS models were induced by E.coli (1 ml/100 g) and a djuvant 10 % BaSO4(1 ml/100 g)abdominal injection.The rats in treatment group w ere administered orally with TFKYD at the same time of the injection,while the r ats in the model group were administered orally with the same amount of saline a t the same time of the injection.The rats in the normal group were injected with the same amount of saline.The conditions of movement, breath, urine and stool w ere observed.The WBC,IL- 1,IL- 6,IL- 10 levels were measured from the venous blood from it′ s tail 24 hours after the injection. The variety of pathologic m orphology of the rat′ s liver,lung,kidney and small intestine were observed und er light microscope and photographed after 48- hour treatment.Result:Condition s of rats in both treatment group and model group degenerated.The WBC, IL- 1, I L- 6,IL- 10 levels of the treatment group and the model group were much higher than those of the normal group. But the WBC, IL- 1, IL- 6 level of the treatm ent group were lower than those of the model group, while the IL- 10 level was higher than that of the model group. The inflammatory lesion of the rat′ s live r,lung,kidney and small intestine of the treatment group were slighter than that of the model group.Conclusion:TFKYD can inhibit or abate the process of infecti ous SIRS /MODS through binary modulation of the cytokines and it can relieve the inflammatory lesion of the organs.
