七氟醚对缺氧无糖损伤大鼠海马NR_1亚基mRNA表达的影响 被引量:1

摘要 目的探讨七氟醚对缺氧无糖(oxygenandglucosedeprivation,OGD)损伤大鼠海马NMDA受体NR1亚基mRNA表达的影响。方法大鼠海马脑片随机分为3组(n=3),用RT-PCR方法检测对照组、缺氧组及七氟醚组大鼠离体海马脑片OGD损伤14min恢复氧糖供应孵育1、2、4h后NMDA受体NR1亚基mRNA的表达。结果恢复氧糖供应1、2h后NR1亚基mRNA的表达3组无明显差异,而缺氧组4h后NR1亚基mRNA的表达增高,七氟醚组恢复氧糖供应后4hNR1亚基mRNA的表达明显降低。结论七氟醚可通过下调OGD损伤引起的NR1亚基mRNA的表达发挥脑保护作用。 Objective To study the effect of sevoflurane on the expression of mRNA of NR_1, one subgroup of NMDA receptor, in rat hippocampal slices deprived of oxygen and glucose(OGD). Methods The rat hippocampal slices were randomly divided into three groups, the normal control group, hypoxic OGD group and sevoflurane treatment group. The slices were expossed to OGD for 14 min and then incubated with oxygen and glucose resupplied for 1, 2 or 4 h. RT-PCR technique was used to study the expression of NR_1 mRNA of NMDA receptor. Results No significant differences in the expression of NR_1 mRNA were found between the groups at the 1 and 2 h time points. However, the expressions of NR_1 mRNA was increased at 4 h in the hypoxic group(P<0.05), but the increase was not noticed in the sevoflurane treatment group(P<0.05). Conclusion The neuroprotection of sevoflurane is possibly attributed to its downregulating the expression of NR_1 mRNA of NMDA receptor after OGD.
