摘要
以凯氏定氮法测定结果为基础,采用紫外分光光度法测定转移因子透析液中的多肽含量。该法的回收率及相对标准偏差分别为99.56%、1.48%(n=6);经与Folin-酚法相比较,确定两种方法测得含量的比值为7.17,RSD为3.77%(n=20)、相关系数为0.93;并推算出当吸收值为15.10时,多肽含量为1.0mg/ml。从而为转移因子透析液中多肽的定量提供了一个快速、简便、准确的方法。
Ultraviolet spectrophotometry(UVS)for the determination of polypeptide in thetransfer factor(TF)extractive was established. The concentration of polypeptide in TF extractivewas first detected with kjeldahl-gunning method for nitrogen determination.Basedon the resultsablained,the standard cuvre of UVS was laid down.The recovery of UVS was 99.56%±1.48%(n=6). The results of UVS were well correlated with those of Folin-phenol method(r=0. 93,n=20). The ratio of the concentrations determined by the two methods,C_(UV)/C_F was 7. 17±3. 77%.Itwas showed that the concentration of polypeptide is 1.0mg/ml when the UV absorbance of TFextractive is 15. 10. This study provides a convenient ,rapid and quantitative method for the deter-mination of TF extractive.
