摘要
目的探讨CuZn超氧化物歧化酶(CuZnSOD)的代谢变化与胃癌组织发生及发展之间的相互关系.方法 52例胃癌患者手术切除的新鲜胃标本,检测胃壁组织中CuZnSOD的活性;应用免疫组化的方法检测CuZnSOD在胃壁组织中的分布,并采用病理图像分析的方法进行灰度检测.结果胃非粘液性腺癌Ⅰ~Ⅳ级癌灶组织中CuZnSOD活性分别为:(62.85±8.47)、(60.94±6.51)、(36.76±3.73)、(39.04±3.85)NU/mgPr;其癌细胞中CuZnSOD灰度分别为:(128.91±14.11)、(120.07±5.34)、(97.73±5.87)、(83.83±7.28).胃粘液性腺癌组织中CuZnSOD活性为(17.73±1.74)NU/mgPr;其癌细胞多不表达CuZnSOD.CuZnSOD在正常对照组胃泌酸腺细胞中呈阳性表达,在胃粘液性腺细胞中呈阴性表达.结论胃非粘液性腺癌的发生可能来源于分布在胃体和胃底的胃泌酸腺细胞,CuZnSOD具有促进其癌细胞分裂和增生的作用.胃粘液性腺癌的发生可能来源于胃粘液性腺细胞,CuZnSOD与其癌细胞的发生、分裂和增生无关.
Objective To investigate the relationship between the metabolic changes of the Cu-Zn-superoxide dismutase (CuZnSOD) in the gastric tissue and the gastric carcinogenesis and development of the gastric carcinoma.Methods In the fresh gastric samples surgically resected from 52 cases of gastric carcinoma,the CuZnSOD activity and distribution was measured by immunohistochemical assay. The gray scale was monitored by means of pathomorphological analysis.Results In the non-mucinous adenocarcinoma of stomach,the CuZnSOD activities in the gastric focus tissue with grades Ⅰ-Ⅳ were (62.85±8.47),(60.94±6.51),(36.76±3.73),(39.04±3.85) NU/mg Pr respectively,and the gray scales in the tumor cells were (128.91±14.11),(120.07±5.34),(97.73±5.87),(83.83±7.28) respectively. In the mucinous adenocarcinoma of stomach,the CuZnSOD activity in the gastric focus tissue was (17.73±1.74) NU/mg Pr,and the expression of CuZnSOD was not detectable in the tumor cells. In normal control group,the expression of CuZnSOD in the gastric oxyntic gland cells and the gastric mucous glandular cells was positive and negative respectively.Conclusion The gastric carcinogenesis of the non-mucinous adenocarcinoma of stomach maybe originate from the gastric oxyntic gland cells in the gastric corpus and fundus,and CuZnSOD in the mucinous adenocarcinoma of stomach could advance the division and proliferation of tumor cells. The gastric carcinogenesis of the mucinous adenocarcinoma of stomach maybe originate from the gastric mucous glandular cells,and CuZnSOD was not related with the carcinogenesis,division and proliferation of the tumor cells.
