摘要
目的应用改良PBS对Y.pseudotubercolosis进行低温增菌和碱处理试验。方法用生理盐水分别稀释假结核耶尔森氏菌521,522,F23株菌,然后分别转种在改良PBS液体内,置4°C增菌于不同时间取出,接着经碱处理后进行分离培养。结果Y.pseudotubercolosis521,522株在21天出现一个峰值,F2株在14天达到高峰后维持至28天不变;碱处理不影响分离率。结论改良PBS低温增菌培养方法简便,经碱处理对其结果没有影响,可应用于Y.pseudotubercolosis增菌培养。
Objective The test of the action of improved phosphate buffer solution(PBS) on Y.pseudotubercolosis through low-temperature bactria-proliferating and alkaline treatment was made. Methods Three strains of 521, 522, F 2 of Y. pseudotubercolosis were diluted with physiological saline respectively and then they were transplanted in improved PBS respectively and placed at 4°C and taken out at different times and isolated for culture after alkaline treatment. Results The strains of 521 and 522 of Y.pseudotubercolosis showed a peak value on 21st day and F 2 strain kept unchanged to 28th day after reaching a peak value on 14th day; there was no difference in detection rate using alkaline treatment. Conclusions The method of low-temperature bacteria-proliferating culture using PBS is simple and convenient. There is no effet of using alkaline treatment on detected results. This test can applied to bacteria-proliferating culture of Y.pseudotubercolosis.
