摘要
在动物实验证明活血化瘀复方血管通具有预防动脉粥样硬化(AS)形成作用的基础上,本实验采用20只新西兰纯系家兔,分成四组,正常组、高脂组、高脂加阿斯匹林组及高脂加血管通组,取兔升主动脉根部组织进行超微结构观察;体外动脉平滑肌细胞(VSMC)培养,测定同步化VSMC^3H—TdR掺入量,进一步研究血管通对VSMC增生的影响,结果显示:血管通可抑制SMC从收缩表型向合成表型转变,并可使SMC对~3H—TdR掺入量显明低于高脂组,抑制动脉SMC的DNA合成,以抑制SMC增生。提示血管通抗AS的作用机制可能与其抑制VSMC有关。
<abstract>The effect of Xue Guan Tong(XGT)on vascular smooth muscle celKVSMOproliferation in atherosclerotic rabbits was studied in order to investigate the mechanism of effect of XGT on inhibiting atherosclerotic formation. Twenty purebred New Zealand rabbits were randomly divided into four groups:normal group, model group,aspirin group and XGT group(6g/kg/d). Ascending aorta of two rabbits in each group was taken for preparation of specimens for transmission election micrograph. The 2cm up part of thoracic aorta was trimmed for VSMC culture and 3H -TdR incorpora-tion assays. The results were shown that XGT could prevent VSMC transformation from contractive stage to synthetic stage and reduce 3H - TdR incorporation, which indicated that XGT could inhibit VSMC DNA synthesis and proliferation. This experiment suggested that XGT had the potentiality to prevent the development of atherosclerosis by inhibiting VSMC proliferation.
