摘要 目的研究Caspase3在N甲基N亚硝脲(MNU)诱导的光感受器细胞凋亡过程中的表达并观察其与细胞凋亡的关系。方法大鼠MNU腹腔注射造模后,分别在不同时间段,利用免疫组织化学法检测视网膜中Caspase3的表达,同时用TUNEL法检测视网膜细胞的凋亡。结果MNU腹腔注射后,外核层中Caspase3的表达渐增强,至2d时达高峰,此后渐下降。细胞凋亡数量的变化趋势与前者一致。结论Caspase3可能在MNU诱导的光感受器细胞凋亡中发挥主要作用,其表达量的高低与细胞凋亡的程度有密切联系。 ObjectiveTo investigate the relationship between Caspase-3 expression and apoptosis in N-methyl-N-nitrosourea (MNU)-induced retinal degeneration model.MethodsMNU was intraperitoneally injected in rats and the experimental rats were killed at scheduled time point during the 12 hours to 5 days.Caspase-3 expression was determined using immunohischemistry and photoreceptor apoptosis was marked with terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) staining.ResultsThe positive expression of Caspase-3 was detected in the outer nuclear layer (ONL) at 12 hours and reached the peak on 2 days,and then the expression level was gradually reduced from 2 to 5 days after MNU injection.The changes of TUNEL-positive cells followed the resemble pattern to the former.ConclusionCaspase-3 may play a important role in the MNU-induced retinal degeneration,and its expression may be closely related to cell apoptosis.
