摘要
目的:研究姜黄素(Curcumin,Cur)对糖尿病大鼠肾组织一氧化氮水平的影响及对肾脏的保护作用。方法:链脲佐菌素(STZ)诱导糖尿病大鼠模型,并随机分为糖尿病组和姜黄素治疗组,另设一正常对照组。治疗组组予姜黄素200mg/(kg·d)灌胃,为期8周。然后观察肾脏结构和功能及肾皮质一氧化氮(NO)水平的变化。结果:与正常对照组大鼠比较,糖尿病组大鼠、尿白蛋白排泄率(UAER)、尿白蛋白/肌酐(Alb/Cr)、肾重(KW)及肾肥大指数(KW/BW)明显升高;光镜示肾小球体积明显增大,肾小球平均截面积(MGA)和肾小球平均体积(MGV)明显增加;肾皮质NO水平明显增加。而姜黄素治疗组上述异常均明显减轻。结论:姜黄素能降低早期糖尿病大鼠肾组织NO水平,减轻肾小球高滤过和肾脏肥大,降低尿白蛋白,改善肾功能。
Objective:To investigate the influence of curcumin on the level of nitric oxide in kidney and the renal protective effect in diabetic rats.Methods:Diabetic models were induced by a single intraperitoneal injection of streptozoticin(STZ)into male Wistar rats at a dose of 60mg/kg body weight and was randomly divided into two groups:untreated diabetes group(group DM,n=10) and diabetes group treated with curcumin 200mg·kg -1 ·d -1 (group Cur,n=10).A normal contral group(group N,n=10)was also designed,After treated for eight weeks,the level of NO in kidney,renal structure and function were evaluated.Results:The levels of urinary albumin excretion rate (UAER),albumin-creatinine ratio(Alb/Cr),kidney weight(KW),profile of kidney hypertrophy(KW/BW),mean glomerular area (MGA) and mean glomerular volume(MGV)were all significantly increased in dialetic rats as compared with normal rats (all P<0.01);larger glomeruli could be found by light microscopy;the level of nitric oxide(NO)in renal cortex of diabetic rats was much higher than that in normal rats.All these abnormalities were markedly ameliorated when treated with curcumin.The levels of blood urea nitrogen(BUN) and serum creatinine(Scr) have no significant difference among the three groups.Conclusion:Curcumin may decrease the level of NO in kidney of diabetic rats at early stage, alleviate kidney hyperfiltration and hypertrophy,and decline UAER.
