用反相高效液相色谱—电化学法测定血中左旋多巴和卡比多巴浓度 被引量:1

摘要 本文用HPLC-电化学法测定血中左旋多巴(Levodopa)和卡比多巴(Carbidopa)浓度,并对家兔体内药动学进行了初步研究。结果表明:两药在25-6000ng/ml范围内,峰高与浓度具有良好的相关性(两药均为r=0.9993),最低检测限均为0.4ng。日内变异系数小于12%,日间变异系数小于15%。回收率左旋多巴为97.67%,卡比多巴为88.55%。家兔口服复方多巴片后,药时曲线为一房室模型,左旋多巴半衰期为2.27h,卡比多巴半衰期为3.22h,与国外文献报道基本一致。用HPLC-电化学法测定血中左旋多巴和卡比多巴脓度,为帕金森氏病的合理用药提供一种稳定、可靠的重要工具。 he plasma concentration of levodopa and carbidopa was determined by Hight-performance liquid chromatography withelectro-chem ical detection simultaneously , and the pharmacokinetic of levodopa and carbidopa in rabbit was studied. The re-sults showed that the concentration range was 25-6000ng/ml(levodopa r= 0.9992, carbidopa r= 0. 9993). The detection lim-it was 20ng/ml. The RSD of within-day and between- day were respectively lower than 12%and 15%,The mean recoverieswere 97.6%(levodopa) and 88. 55%(carbidopa).An open model of one compartments was the best approach for the phar-macokinetic profile of levedopa and carbidopa administered orally to the rabbits. The biological half-lifes (t1/2)of levedopaand carbidopa respoctively 2. 27h and 3. 22 h , it was similar to external references. The determination of levodopa and car-bidopa in plasma offered a sensitive reliable assay method for the treatment of Parkinson' s disease.
