聚乙烯颗粒诱导假体周围骨溶解中TNF—α变化的实验研究 被引量:3

摘要 目的 了解TNF-α在聚乙烯颗粒诱导假体周围骨溶解中的作用及其机理。方法 将钛合金棒植入大鼠双侧股骨远段,2周后向左侧膝关节注射聚乙烯颗粒,右侧注射生理盐水对照,每2周注射一次,12周时处死动物,取假体周围界膜组织和膝关节滑膜作组织学观察,ELISA法测定假体周围界膜组织TNF-α的浓度。结果 注射聚乙烯颗粒侧关节滑膜及假体周围界膜组织可见明显的异物肉芽肿反应,假体周围界膜组织TNF-α含量明显增高。结论 聚乙烯颗粒能引起假体周围骨溶解,TNF-α可能起着重要作用。 Objective To investigate the effect of TNF-α in periprosthetic osteolysis induced by polyethylene particles. Methods Titanium alloy plug was inserted into each distal femur through the knee. After two weeks, polyethylene particles were injected into the left knee and saline solution was injected into the right knee as the control every two weeks. The animals were killed after twelve weeks. The periprosthetic interface membrane and joint synovium were obtained to observe the histopathological changes. The concentration of TNF-α in membrane was assayed by ELISA kit. Results A large number of macrophages, fibroblasts and multi-nuclear giant cells were seen in the synovium of knee and the membrane around the plug on the left side. The concentration of TNF-α in the periprosthetic interface membrane on the left side was higher than right side. Conclusion Polyethylene particles could induce the inflammatory response around the Titanium alloy plug and increase the secretion of TNF-α, which may play an important role in the periprosthetic osteolysis induced by wear debris.
