Effect of Simvastatin on Myocardial Oxidative Stress during Decompensated Right Heart Failure in Rats

摘要 目的:观察辛伐他汀(Sim)对野百合碱(MCT)引起的失代偿右心衰竭大鼠心肌的氧化应激的影响。方法:一次性皮下注射MCT制备大鼠右心衰竭模型。注射MCT两周后,灌胃给予Sim或蒸馏水(对照组)治疗14d。测定各组大鼠右室心肌匀浆中的SOD和GSH-PX酶活力及MDA含量,以及肝和肺组织的湿重和干重的比值。结果:与正常大鼠比较,对照组大鼠右室心肌SOD和GSH-PX酶活力显著下降,心肌MDA含量及肝、肺湿重和干重的比值明显升高,(P<0.01)。Sim可有效地抑制MCT引起的心肌SOD和GSH-PX酶活力的下降及MDA含量的增加(P<0.01),并使肝、肺湿重和干重的比值降低。结论:Sim对MCT引起的失代偿右心衰竭大鼠心肌的氧化应激具有抑制作用。 Objective: To observe the effect of simvastatin(Sim) on myocardial oxidative stress during decompensated right heart failure induced by monocrotaline(MCT) in rats.Method: Rats were given a single subcutaneous injection of MCT to produce right heart failure. Two weeks after injection of MCT, rats were treated with Sim or distilled water (control) for fourteen days. Activities of SOD and GSH-PX and content of MDA in the right ventricular homogenate and wet to dry weight ratio in the lung and liver were measured. Results: In control group , the SOD and GSH-PX activities decreased and the MDA content was increased in the myocardial, and the ratio of wet to dry weight in the liver and lung was increased, compared with normal (P<0.01). Sim can effectively inhibit the decrease of SOD and GSH-PX activities, and the increase of MDA content in the myocardial, and the increase of wet to dry weight ratio in the liver and lung, induced by MCT, compared with control (P<0.01). Conclusion: Sim can inhibit the myocardial oxidative stress during decompensated right heart failure induced by MCT in rats.
