年龄相关性白内障晶体上皮细胞凋亡与端粒酶的表达 被引量:1

摘要 目的:探讨年龄相关性白内障及正常晶状体上皮细胞的端粒酶活性的表达及其在年龄相关性白内障发生发展中的作用。方法:实验于2003-01/06在福建医科大学附属第一医院中心实验室完成。采用端粒重复序列扩增-银染法及端粒重复序列扩增-酶联免疫吸附法两种方法检测9例年龄相关性白内障、3例正常晶状体上皮细胞的端粒酶活性。结果:端粒重复序列扩增-酶联免疫吸附法检测结果:9例年龄相关性白内障及3例正常晶状体上皮细胞中端粒酶均呈阳性,A值分别为0.5371±0.3407,1.2077±0.1545,年龄相关性白内障组晶状体上皮细胞中端粒酶活性明显低于正常组,差异具有显著性意义(χ2=66.23,P<0.05)。端粒重复序列扩增-银染法检测结果:阳性对照正常、年龄相关性白内障晶状体上皮细胞标本的提取液均为阳性,可见52bp及相差6bp的梯性条带,阴性对照标本均未见梯性条带。结论:端粒酶活性的降低可能是年龄相关性白内障晶状体上皮细胞发生凋亡的一个重要机制,端粒酶在年龄相关性白内障的发生发展中起着重要的作用。 AIM:To investigate the expression of telomerase activity in human lens epithe lial cells of age- related cataract and normal lens,and evaluate the role of te lomerase activity in the occurrence and progression of age- related cataract. METHODS:The experiment was conducted in the central laboratory in the First A ffiliated Hospital of Fujian Medical University from January to June 2003.Telome re repeat amplificaton protocol(TRAP) sliver staining method and TRAP- enzyme- linked immunoadsordent assay(ELISA) method were used to detect the telomerase a ctivity in epithelial cells of 9 age- related cataract lens and 3 normal lens. RESULTS:As shown by TRAP- ELISA dectection,telomerase activity in lens epith elial cells of the 9 age- related cataract and the 3 normal lens was all positi ve,A value was 0.537 1± 0.340 7 and 1.207 7± 0.154 5,respectively,indicating t hat the telomerase activity in the lens epithelial cells of the former were sign ificantly lower than that of the latter(χ 2=66.23,P< 0.05).From the detection o f TRAP silver staining,the liquid extracted from the epithelial cell specimens o f the normal lens and age- related cataract was all positive,and 52 bp and phas e difference 6 bp ladder band were found in them,but no band was found in the ne gative control speciments. CONCLUSION:The decrease of telomerase activity may be an important mechanism of apoptosis of age- related cataract lens epithelial cells,indicating that tel omerase plays an important role in the occurrence and progression of age- relat ed cataract.
