CyclinD1、PCNA、Ki67细胞周期相关蛋白在喉复发癌组织芯片中的表达和意义

Significance of Cyclin D1, PCNA, and Ki 67 protein expressions in tissue arrays of patients with recurrent laryngeal carcinoma

摘要 目的研究Cyclin D1、PCNA、Ki-67细胞周期相关蛋白的表达与喉癌复发的关系,并评价其对预后评估的作用.方法回顾分析1958~1999年喉癌手术病例574例,术后随访3年,确诊术后复发56例;正常喉对照标本39例.标本制成组织芯片,采用免疫组化SABC法,对芯片标本进行染色.结果 Cyclin D1、PCNA、Ki-67在喉癌组织及正常喉组织中均有表达,且在喉癌组织的表达强于正常喉组织(P<0.01),在喉癌复发组强于未复发组(P<0.01),与喉癌复发呈正相关.结论 Cyclin D1、PCNA、Ki-67细胞周期相关蛋白的表达可作为估计喉癌预后的指标. Objective To evaluate the prognosis of laryngeal cancer postoperative recurrence, and to investigate the relationship of Cyclin D1, PCNA, and Ki 67 protein expression in patients with recurrent laryngeal carcinoma. Methods 574 cases of laryngeal carcinoma from 1958 to 1999 were retrospectively reviewed. All patients were follow up surveyed for 3 years postoperatively. The whole set of tissue microarrays from 574 patients contained tumor tissue, including recurrent tumor tissue from 56 patients, and 39 normal laryngeal tissue specimens from normal laryngeal were collected as control. The tissue chips were analyzed by the SABC immunohistochemical method. Results Cyclin D1, PCNA, and Ki 67 expressed in most of the laryngeal carcinoma and normal laryngeal tissues in a different positive ratio. There was higher expression of Cyclin D1, PCNA, and Ki 67 in recurrent laryngeal carcinoma compared to that in laryngeal carcinoma without recurrence and normal laryngeal tissue ( P <0 01). There was positive correlation between the expression of Cyclin D1, PCNA, and Ki 67 and the recurrent laryngeal carcinoma. The expression of those genes was also significantly different between in the laryngeal carcinoma and in normal laryngeal ( P <0 01). Conclusion Cyclin D1, PCNA, and Ki 67 were the prognostic marker candidates for the postoperative laryngeal cancer recurrence
