青蒿琥酯对小鼠曼氏血吸虫虫卵肉芽肿形成的影响 被引量:5

摘要 目的通过口服青蒿琥酯,观察其对曼氏血吸虫感染小鼠肝脏肉芽肿形成的影响。方法小鼠经尾部接触感染曼氏血吸虫尾蚴后,经不同时间、不同剂量喂服青蒿琥酯,治疗后2~6周解剖,取肝脏作石蜡切片,进行H.E.染色,显微镜下作病理学观察,并测定虫卵肉芽肿的面积。结果未给药对照组虫卵肉芽肿由大量炎性浸润细胞及间质成纤维细胞组成,服药组肝切片未见虫卵肉芽肿或虫卵肉芽肿面积明显小于对照组,肉芽肿周围少量炎性细胞浸润。100mg/kg组肉芽肿面积与对照比较无显著性差异(P>0.05)。但肉芽肿面积比对照减小36.9%。300、500mg/kg组病理切片未查到虫卵和肉芽肿。不同虫龄服药组肉芽肿面积与对照比较均有显著性差异(P<0.05~P<0.01)。其中21、22d服药组肝切片未查到虫卵和肉芽肿。结论青蒿琥酯能抑制虫卵肉芽肿的形成,降低虫体产卵率并减少形成的肉芽肿对肝脏的损害。 To observe the effect of Artesunate on the egg granuloma formation of Schistosoma mansoni, BALB/c mice infected with S.mansoni cercariae by tail-dipping method were administrated orally with Artesunate at different dosages and at different developmental stages of parasites. After 2-6 weeks of treatment, mice were scarified, and the paraffin-embedded sections of liver were stained by hematoxylin-eosin(HE),and underwent the pathological examination. The size of granulomas was measured by using a VM-30 videomicrometer and NIH image software for more than 10 granulomas in each mouse. And the average granuloma area (μm^2) was calculated for each mouse. It was found that most granuloma in liver of mice in the control group consisted of numerous infiltrations with inflammatory cells and mesenchymal fibroblast, while in the treated group of mice, no granuloma formation was observed or the size of granuloma was significantly reduced in comparison with that of the control group. And there were only small amount of infiltrations with inflammatory cells around the granulomas at dosage of 100 mg/kg Artesunate administrated, no significant difference between the treated and control groups was observed, but the average size of egg-granuloma formation was reduced by 36.9%, but at dosages of 300 mg/kg and 500 mg/kg, no egg was found in the pathological sections or the egg-granuloma could be observed . At different developmental stages of parasites, the size of egg-granulomas in liver of untreated group was larger than that of the treated group of mice with significant differences. Also, no egg or egg-granuloma formation could be found in the 21 days or 22 days development of parasites. It is concluded that Artesunate can inhibit the egg-granuloma formation of S.mansoni, decrease the rate of egg production,and reduce the damages in liver induced by the guanuloma formation.
