摘要
目的TGFβ1疫苗诱导Balb/c小鼠产生抗体及对TGFβ1体外中和活性。方法通过已构建成功的TGFβ1疫苗免疫小鼠,诱导产生高效价中和抗体,抗TGFβ1抗体及合成的抗原肽ELISA法检测免疫血清抗体滴度,经貂肺上皮细胞生长抑制法,检测抗体血清对TGFβ1体外中和活性。结果抗TGFβ1疫苗可以诱导高滴度的抗体产生,可与抗TGF抗体及抗原肽结合,可以阻断TGFβ1对貂肺上皮细胞的抑制活性。结论构建的抗TGFβ1疫苗可以诱导高滴度的中和抗体产生,在体外阻断TGFβ1活性,可以作为抗纤维化疫苗用于抗纤维化动物模型研究。
Objective MTT methods detect the antibody neutralization activity induced by TGF-β_1 vaccine. Methods The human TGF-β_1 epitope inserted the elloop of C-te rminus of truncated hepatitis B core antigen as a fusion protein immunized BALB/ c mice as vaccine the antigenicity of recombinant antigen was determined by ELIS A.The serum antibody was detected by TGF-β 1~)32( antigenic peptide.TGF -β_1 activity was measured by MTT using epithelial cells Mv1Lu as inhibiting cell s. Results The fusion protein had the TGF-β_1 antigenicity but hadn't HBc antigenicity.A high anti-TGF β_1 response and weak anti-HBc respo nse were observed in mice after immunized,the highest end point dilution titers of anti-TGF β_1 and anti-HBc were 1∶3.2×10~5 and 1∶1.6×10~3 respective ly.The serum antibody can prevent TGF β_1 activity. Conclusion Recombinant fusion protein can be a cytokine vaccine t o induced high titers anti-TGF-β_1 antibody.The results emphasize the potent ial for fusion proteins can as vaccines to prevent liver fibrosis.
