摘要
目的:观察抑癌基因Tip30联合紫杉醇抑制腺样囊性癌细胞ACC-2生长及特征性细胞核DNA变化。方法:待检ACC2细胞分为4组:紫杉醇组、转基因组、紫杉醇+转基因组、空白对照组,采用MTT法检测细胞增殖率;DNA片段分析检测DNA裂解。结果:不同处理组检测结果分析显示,紫杉醇+Tip30组ACC2细胞生长明显受到抑制,DNA裂解片段最为明显;单项处理组细胞生长亦受到抑制;对照组细胞生长未受影响。结论:Tip30联合紫杉醇可显著抑制ACC-2细胞生长,出现特征性DNAladder现象。
Objective:To study the inhibitory effects of Tip30 combined with paclitaxel on the growth of salivary gland adenoid cystic carcinoma cells(ACC-2) and observe the characteristic changes of nuclear DNA of ACC-2 cells.Methods: ACC-2 cells were divided into 4 groups: paclitaxel group, pcDNA-Tip30 transfecting group, paclitaxel+pcDNA-Tip30 transfecting group, and control group. MTT assay was used for detecting cell proliferation, and DNA fragment analysis was used for DNA cleavage in 4 groups. Results: The growth of ACC-2 cells in Paclitaxel+Tip30 transfecting group was inhibited significantly with obvious DNA fragments. The cell growth in paclitaxel group and pcDNA-Tip30 transfecting group was also inhibited, but cell growth in control group was not affected. Conclusion: Tip30 combined with Paclitaxel can obviously inhibit the growth of ACC-2 cells and cause the characteristic DNA ladder phenomenon.
