补肾壮筋汤对实验性骨关节病NO和软骨细胞凋亡的影响 被引量:15

摘要 目的:探讨补肾壮筋汤对兔膝骨关节病的血清和关节液一氧化氮(nitricoxide,NO)含量,软骨细胞凋亡和增殖的影响及其内在关系。方法:青紫蓝兔,50只分5组,参照Hulth’s法造模。7周末测定血清和膝关节液NO值;光镜,透射电镜下观察软骨形态学变化;免疫组化法观察软骨细胞凋亡和增殖情况。结果:血清和关节液NO值有相关性(P<0.01),模型组NO值和其他所有组比较有统计学意义(P<0.05);中药组的凋亡细胞指数、增殖指数与模型组比较有统计学意义(P<0.05)。结论:补肾壮筋汤能降低兔膝骨关节病血清和关节液的NO的浓度,减少软骨细胞的凋亡,促进软骨细胞的增殖。 Objective To discuss the changes of NO in serum and Knee joint fluid of experimental osteoarthritis(OA) in rabbits and relation between them by Bu Shen Zhuang Jin Decoction,and to observe the effects of BuShenZhangJin Decoction on apoptosis and the expression of proliferating cell nuclear antigen(PCNA)of chondrocytes Methods OA models were established with hulth's method.The NO contents of serum and knee joint fluid were examined at the end of 7 weeks.The changes of morphology of chondrocyte were observed by light microscopy and transmission electron microscopy and examined by immunohistochemistry for PCNA and TUNEL for apoptosis.Results There was significant relation of NO between serum and knee joint fluid.Compared with others,NO of the model group had statistically significant differences(P<0.05).Apoptosis cell index and cell proliferation index of BuShenZhuangJin Decoction group had significant differences too (P<0.05). Conclusion BuShenZhuangJin Decoction could decrease the NO in serum and knee joint fluid of experimental osteoarthritis in rabbits,decrease the apoptosis of chondrocytes,increase chondrocyte proliferation,showing therapeutic effect to experimental osteoarthritis.
