干细胞凝胶复合体加右归饮修复家兔软骨缺损的实验研究 被引量:16

摘要 为探讨右归饮对骨髓基质干细胞凝胶复合体修复兔关节软骨缺损的影响。取兔骨髓基质干细胞进行体外增殖后,复合于纤维蛋白凝胶,并加入细胞因子TGFβ与IGFⅠ,植入到兔膝关节实验性关节软骨缺损,并以中药右归饮灌胃。4、8、12周后对修复组织进行形态学观察及组织学检查。结果术后12周,右归饮实验组的缺损由透明软骨样组织充填,软骨及软骨下骨组织基本修复,修复的软骨组织在组织形态学上优于对照组。表明骨髓基质干细胞+纤维蛋白凝胶+TGFβ、IGFⅠ能修复家兔膝关节软骨缺损,中药右归饮能促进软骨的修复,且提高修复质量。 The objective of the paper is to explore the influence of 'YOUGUI LING'(YGY, the Kidney-Yang-Reinforcing Decoction) and stem-cell gel complex on the repair of rabbit articular cartilage defect. After the in-vitro culture and multiplication, rabbit spinal stromal stem-cells were mixed with, and TGF-β and IGF-I were added to fibrin gel that stem-cell gel complex was made; then the complex was implanted into experimental rabbit cartilage defect; and the rabbits were given YGY through the stomach. Four, eight and twelve weeks later, the repaired tissues were observed by histology and histomorphology. The results showed that 12 weeks later, the defects of the YGY-experimental group were filled with the hyaline chondroid tissues; the cartilage and subchondral tissues were basically repaired, having an advantage over those of the control in the repaired tissues hisomorphology, suggesting that spinal stromal stem-cells + fibrin gel + TGF-β and IGF-I functioned to repair rabbit knee cartilage defect and TCD YGY could accelerate the cartilage repair and improve the repair quality.
