摘要
目的建立测定人血浆中西替利嗪浓度的高效液相色谱法。方法HypersilBDS-C18(150mm×5.0mm,5μm)色谱柱上,乙腈-0.05mol·L-1磷酸二氢铵(37∶70)为流动相,检测波长231nm,柱温30℃。结果西替利嗪在25~1.0×104μg·L-1范围内线性关系良好(r=0.9988),最低检测浓度为25μg·L-1。低、中、高3种浓度的回收率为99.4%~104.8%,日内、日间相对标准差为2.9%~9.6%。结论所用方法检测西替利嗪的血药浓度,结果准确可靠,可满足临床药物动力学研究的要求。
OBJECTIVE To establish an HPLC method for the determination of cetirizine in human plasma. METHODS The analysis was performed with a Hypersil BDS-C_~18 (150 mm×5.0 mm,5 μm) column, acetonitrile: 0.05 mol·L^-1 ammonium dihydrogen phosphate (37∶70)was served as mobile phase and detective wavelengh at 231 nm. RESULTS The linear range of cetirizine hydrochloride was 25-~1.0×104 μg·L^-1 (r=0.9988)and the average recovery was 99.4%-104.8% at three different concentrations. CONCLUSION This method is suitable for the pharmacokinetic study of cetirizine.
