本文采用紫外分光光度法测定了14例用环磷酰胺治疗的恶性淋巴瘤患者体内的血浆药物浓度,并依据AIC差别程序按静推二室开放模型拟合后进行非线性迭代回归计算药代动力学参数,结果表明:CTX在体内以较快的速度进行分布,经过24h左右药物几乎全部被消除;主要参数为:t3/2α13.79±11.90min,t1/2β339.73±204.27min,CL1.453×10^2ml/min,AUC(9.
A sensitive colorimeter technic is described for determining cydophosphamide(CTX)in plasma. It isbased on the fact that active alkylating metabolites of CTX became purple in color by absorbing 575nm at basic pH. The minimal amount of detection is 1. 5ug/ml in plasma. Plasma level of CTX in 14 patients with Lymphoma had been mea-sured after i. v. dosage of 600-800mg/m2.of CTX. The concentration-time curve fitted is well with the compartment open model. The concentration-time curve was calulated with non-linear iteration regression by computer. The pharmacokinetic parameters showed that tl/2α, tl/2β,CL, AUC and Vd were 13. 79min, 339. 73min, 1. 453 × 102ml/min, 9. 554 × 104ng/ml min and 0. 89L/kg respectively.
