摘要
目的观察川芎嗪和尼可地尔对兔急性心肌梗死再灌注后心肌“无复流”现象的影响,并将二者进行对比观察。方法新西兰大白兔45只,随机分为川芎嗪组(Ⅰ组)、尼可地尔组(Ⅱ组)、生理盐水对照组(Ⅲ组),每组15只。结扎冠状动脉左心室后支90min,切断结扎线后各组分别静脉滴注川芎嗪、尼可地尔和生理盐水。观察指标:①左心室的心肌无复流面积(NRA/LVA):②左心室心肌梗死面积(MIA/LVA);③血清心肌磷酸肌酸激酶(CK)、乳酸脱氢酶(LDH)、超氧化物岐化酶(SOD)和丙二醛(MDA)。结果Ⅰ组和Ⅱ组的NRA/LVA和MIA/LVA均小于Ⅲ组,再灌注后120min时CK、LDH和MDA均低于Ⅲ组;而SOD均高于Ⅲ组。Ⅰ组与Ⅱ组间比较,除了Ⅰ组的CK和LDH低于Ⅱ组外,其他观察指标均无显著差异。结论川芎嗪可减少兔心肌梗死再灌注后心肌无复流面积和心肌梗死面积,减轻心肌损伤,且效果与尼可地尔相当。
Objective To explore the effects of liqustrazin contrasted with nicorandilon the no-reflow phenomenon in rabbits undergoing acute coronary artery occlusion-reperfusion.Methods 45 New Zealand white rabbits were randomly assigned to one of the following three groups:group Ⅰ(ligustrazine),group Ⅱ(nicorandil),group Ⅲ (NaCl saltsolution,NS).Rabbit's acute ischemia-reperfusion model was carried out by ligation of left post coronary descending artery and removing the ligation after occlusion of blood flow for 90 minutes,followed by 120 minutes of reperfusion.Ligustrazinwe,nicorandil and 0.9% NaCl solution (NS) were given intravenously,respectively.The serum levels of CK,LDH,MDA and SOD were observed following acute ischemia and reperfusion.The no-reflow area (NRA) and the myocardial infarct area (MIA) were determined by thioflavin S and TTC staining,respectively.Results Ligustrazine and nicorandil both resulted in a significant reduction in NRA/LVA and MIA/LVA,serum levels of CK,LDH and MDA,while increasing the serum level of SOD,in comparison with control animals. Comparing with nicorandil,ligustrazine resulted in a significant decrease in CK and LDH,while there was no significant difference in the effects on NRA/LVA,MIA/LVA,MDA,and SOD between the two groups.Conclusions Ligustrazine can improve no-reflow phenomenon and alleviate myocardial injury in rabbits with experimental acute coronary artery occlusion-reperfusion,its effects are similar to those of nicorandil.
