人类端粒酶逆转录酶基因的反义寡核苷酸对HeLa细胞的端粒酶活性的研究

摘要 目的:研究人类端粒酶逆转录酶RNA(hTR)的反义寡核苷酸对HeLa细胞端粒酶活性的影响。方法:采用PCRELISA法检测HeLa细胞在反义寡核甘酸处理前后端粒酶活性的变化。结果:经反义寡核苷酸处理后,HeLa细胞的生长受到抑制,端粒酶活性明显降低。结论:hTR的反义寡核苷酸显著抑制肿瘤细胞的端粒酶活性,为恶性肿瘤治疗提供了一条新的有效的途径。 Objective:To investigate effects to telomerase activity by antisense oligonnucle otides of human telomerase reverse transcriptase on HeLa cells in culture.Method s: MTT assay was used to investigate the inhibitory effect of antisense oligonnu cleotides on HeLa cell in culture. The telomerase activity in cells treated by a ntisense oligonnucleotides was detected by PCR-ELISA assay which is based upon PCR amplification of the initial telomerase product and detected by ELISA. Resul ts: The growth of HeLa cells was inhibited by antisense oligonnucleotides and t he telomerase activity in Hela cells was significantly reduced. Conclusion: T he antisense oligonnucleotides could directly inhibit the growth of HeLa cells. The mechanism of anti-tumor by antisense oligonnucleotides may be explained by t he inhibition of telomerase activity in cells.
