穿心莲提取物体外抗呼吸道合胞病毒作用的研究 被引量:11

摘要 目的:了解穿心莲提取物对呼吸道合胞病毒(RSV)的体外抑制作用。方法:通过光镜观察不同药物浓度下RSV对Hep2的致病作用(CPE),四甲基偶氮唑盐(MTT)比色检测穿心莲提取物对RSV的抑制和对RSV感染Hep2细胞的保护作用。结果:穿心莲提取物对RSV无直接灭活作用,亦不能阻止RSV的吸附,各药物浓度均出现典型的CPE;穿心莲提取物有抑制RSV生物合成的作用,其半数抑制浓度(IC50)是1.52μg/ml,治疗指数为36,且随着药物浓度的增加,CPE特征明显逐渐减弱,病毒滴度逐渐降低,而病毒抑制率则明显增高。结论:穿心莲提取物在体外可能是通过抑制生物合成来发挥抗RSV作用。 Objective: To explore anti-respiratory syncytical virus effect of common andrographis her b in vitro. Methods: Microscope was used to observe the effect of common androgr aphis herb on cytopathic effect (CPE), and MTT colorimetric assay were used to d etect the anti-RSV activity and protective function for RSV infected Hep-2 cel ls of common andrographis herb. Results: As typical CPE were observed at all co ncentrations, common andrographis herb had no antiviral abilities of directly ki lling and adsorption blocking. In the group of anti-biological synthesis, the m edian inhibitory concentration (IC50) of common andrographis herb anti- RSV was 1.52μg/mL; the treatment index (TI) was figured out as 36 regarding a median toxic concentration (TC50) of 54.8μg/mL. With the enhancement of the dose, the degree of CPE, viral titers decreased correspondingly, whereas vi ral inhibition rate increased. Conclusion: common andrographis herb may inhibit RSV biological synthesis other than directly inactivate these viruses or block their adsorptions to susceptible cells in vitro.
