藤蟾方抑制人胃癌细胞的生长及诱导其凋亡的实验研究

目的探讨中药复方藤蟾方在体外对人胃癌细胞SGC-7901的生长抑制及凋亡诱导作用。方法用MTT法及流式细胞仪分析法分析藤蟾方对人胃癌细胞SGC-7901细胞的抑制作用及诱导凋亡情况。结果(1)藤蟾方在3.5μg/ml时显示对人胃癌SGC-7901细胞的抑制作用,并随浓度升高而增强,具有显著性差异(P<0.01);(2)通过散点图可看出,随浓度的增加右下象限和右上象限出现高染现象,呈剂量依赖性。其中与空白组比较,低、中、高三剂量的凋亡率均有显著性差异(P<0.01),中、高剂量的坏死率亦有差异(P<0.01或P<0.05)。结论提示藤蟾方在体外能抑制人胃癌细胞SGC-7901的生长,诱导细胞凋亡,这可能为藤蟾方抑制其生长的机理之一。

Objective To discuss the effect of Tengchan recipe on inhibiting growth and inducing apoptosis of human gastric cancer cell SGC-7901 in vitro. Methods Analyze the effect of Tengchan recipe on inhibiting growth and inducing apoptosis of human gastric cancer cell SGC-7901 by MTT and flow cytometry. Results (1) Tengchan recipe inhibit growth of human gastric cancer cell SGC-7901 at 3.5μg/ml and the effect depend on concentration. There are significant difference(P<0.01); (2)The flow cytometry result showed that the effect of inducing apoptosis depended on dose. The effect are significant difference between low, midst, high groups and control group(P<0.01); the necrosis ratio are significant difference between midst group and high group(P<0.01 or P<0.05). Conclusion Tengchan recipe can inhibit growth and induce apoptosis of human gastric cancer cell SGC-7901 in vitro, which may be one of effective mechanisms of Tengchan recipe.
