摘要
以流行性出血热病毒(EHFV)们的组织培养技术,对临床用作EHF抗病毒治疗的各种药物进行了体外抗病毒作用的筛选。结果发现,人白细胞干扰素、聚肌胞、阿糖胞苷、强力宁和华赡素等,既不能阻止EHFV侵入细胞,对细胞内的EHFV增殖也毫无影响。野鼠型免疫血清能中和野鼠型和家鼠型两株病毒,而家鼠型免疫血清只能中和同型病毒。病毒唑25μg/mn或重组干扰素4万IU/ml能有效地抑制细胞内EHFV的增殖,用药愈早,抑制作用愈强。作者还发现,联合病毒唑和重组干扰素具有协同作用,为临床进行抗病毒治疗提出了新设想。
everal drugs have been widely used clinically as antiviral agents in the treatmentof epidemic hemorrhagic fever(EHF).he actual inhibitory effect of each drug on EHFVirus(EHFV)was studied in Vero E6 cell culture inoculated with EHFV- A9 and R22strains ,respectively. They were evaluated on the basis of observing the cytopathic effectas well as testing the viral antigen inside of the infected cells by indirect immunofluo-rescence assay. The results showed that interferon from human leucocytes, poly I :C,ara-C, potenlini and bufonis were not able to influence the replication of EHFV in cellculture or to prevent Vero E6 cells from being infected.Immune serum A16 could neut-ralize both strains before inoculating,but immune serum R22 could neutralize R22 strain only. Intracellular EHFV replication was obviously inhibited by ribavirin 25μg/mlor recombinant interferon 40 000IU/mL,and the earlier they were added ,the strongerthe viruses were affected.It was also found that combining ribavirin and recombinantinterferon in less doses had significant synergic effect on EHFV. These data suggest that the combination of ribavirin and recombinant interferon maybe worthy to be chosen as an effective antiviral regimen in the treatment of EHF, andthe immune serum,if given at early stage,may also be helpful to reduce the sevority ofthis disease.
