摘要
应用正常人骨髓粒细胞系作体外培养并感染流行性出血热病毒(EHFV),分别进行细胞膜脂流动性、扫描电镜和免疫用镜检查。结果发现EHFV感染粒细胞系后1小时开始,感染组细胞膜脂流动性始终低于对照组。扫描电镜发现EHFV感染粒细胞后3天,细胞表面出现凹陷,感染1周细胞膜出现空洞和缺损,并可见细胞肿胀,而对照组细胞均完整。免疫电镜检查发现胶体金颗粒主要沉着在细胞膜上,次为细胞浆内,说明EHFV主要在胞浆内复制,抗原主要在细胞膜上表达。以上结果说明EHFV感染髓细胞后最早引起的是细胞膜病变和损害。
Using the membrane fluidity,scanning electron microscope and immune electronmicroscope technique,we studied the mechanism of cytopathogenic effect of epidemichemorrhagic fever virus(EHFV)on normal human bone marrow cells (HBC).The resultshowcd that membrane fluidity of HBC was significantly lower than that of the con-trol group.Cell membrane appear injured and the major expression of EHFV antigen wason the cell mcmbrane by immune electron microscope. It was suggested that EHFV invadedmainly the cell membrane of HBC,produced damage to the membrane and thereby lesionof the organ cell.
