Optimization of Carbon Sources and Nitrogen Sources in Ferment Culture Medium of Bacterium Producing α-acetolactate Decarboxylase

摘要 对枯草芽孢杆菌3226_5进行碳源、氮源的优化.分别选取单糖、二糖、低聚糖、多糖等7种碳源,以及有机氮和无机氮等9种氮源进行单因子实验,并通过正交试验优化出碳源、氮源的最佳组合.结果表明,以25g/LC2为碳源,12.5g/LN1,7.5g/LN2,5g/LN5,8g/LN酸为氮源的培养基,酶活比原培养基提高了2.73倍. The optimization of carbon and nitrogen sources was made for Bacillus subtilis3226-5 as native strain which could produce α-acetolactate decarboxylase with high efficiency and stability. Seven kinds of sugars including monosaccharide, disaccharide, digosoccharide and polysaccharide, and nine kinds of nitrogen sources including organic nitrogen, inorganic nitrogen were chosen to design single factorial and orthogonal experiments. The result showed that using 25 g/L C_2 as carbon source, 12.5 g/L N_1, 7.5 g/L N_2, 5 g/L N_5 and 8 g/L N_(acidity) as nitrogen sources in the fermentation medium increased the enzyme activity by 2.73 times.
