摘要
旨在进一步观察烧伤后血清触珠蛋白(HP)对B淋巴细胞功能的影响,以期弄清血清HP在烧伤后免疫功能紊乱发生机制中的作用。先采用亲和层析法从烧伤血清纯化HP,然后在培养的正常小鼠脾细胞中分别按正常血清HP含量(火箭电泳法:1.233mg/ml)和烧伤血清HP含量(火箭电泳法:9.407mg/ml)加入纯化HP,观察HP对美洲商陆(PWM)和脂多糖(LPS)诱导的脾细胞IgG产生能力的影响,用ELISA检测培养细胞上清IgG含量。结果表明,相当于烧伤血清HP含量的纯化HP对LPS诱导的脾细胞IgG产生无明显影响,却显著抑制PWM诱导的脾细胞IgG产生;而相当于正常血清HP含量的纯化HP对PWM和LPS诱导的脾细胞IgG产生均无明显影响。提示,烧伤后血清高浓度的HP可能通过抑制T淋巴细胞和/或其淋巴因子的功能而间接影响B淋巴细胞,从而参与烧伤后免疫功能紊乱的发生与发展。

clarified, but it is well known that serum immunosuppressivefactors play an important role in the
pathogenesis of the disturbance of immune response following burn injury. In our
previousstudy, it was found that serum haptoglobin exhibited severe immunosuppressive effect
on T-lymphocyte function following burns, sothis study was intended to investigate the effect of
haptoglohbin purified from the serum of burned patients by affinity chromatogra-phy,thus
clarifying to the role of serum haptoglobin in postburn B-lymphocyte dysfunction. The results
showed that purified hap-toglobin, at the level equivalent to the concentration found in serum of
burned patients, significantly inhibited immunoglobulin G(IgG) synthesis induced by pokeweed
mitogen (PWM),but did not affect IgG synthesis induced by lipopolysaccharide (LPS). Fromthese
result, it is suggested that extraordinary increase in serum haptoglobin following burn may play
a role in the pathogenesis ofB-lymphocyte dysfuntion by suppressing T-lymphocyte function.
