摘要
大肠杆菌O96血清型的大肠杆菌近年来频繁地出现于分离到的致病菌株中,在发展中国家和发达国家均有发现,有造成爆发性流行的可能.本文利用鸟枪法对大肠杆菌(O96O-抗原基因族进行测序,序列全长9415bp,用生物信息学的方法进行序列分析,共发现7个基因,分别是:吡喃型UDP-半乳糖变位酶基因(glf),O-抗原转运酶基因(wzx),O-抗原聚合酶基因(wzy)和糖基转移酶基因(4个).本文分析了大肠杆菌O96和K12(O16)的0-抗原基因族中吡喃型UDP-半乳糖变位酶基因和O-抗原转运酶基因的进化关系,确定了吡喃型UDP-半乳糖变位酶的基序;用PCR的方法筛选出了2个针对大肠杆菌O96的特异基因,可以用于基因芯片或PCR方法快速检测大肠杆菌O96.
The escherichia coli O96 O-antigen gene cluster was sequenced. It was found to contain the genes for UDP-galactopyranose mutase (glf), O-unit flippase (wzx ), O-antigenpolymerase (wzy) and putative transferases. We proposed the genes glf and wzx were evolutionarily related to those of E. coli K12 (O16). We also proposed the motif for UDP-galactopyranose mutase. By polymerase chain reaction against representative stains for the all 166 E. coli and 43 Shigella O serotypes, we identified 2 genes that are highly specific to E. coli O96. This work provides the basis for a quick and sensitive test of the rapid detection of E. coli O96 by DNA array and PCR method.
