摘要
目的建立血浆中阿克他利(Actarit,Acta)的高效液相色谱检测法.方法采用高效液相色谱联用紫外检测器,检测血浆中阿克他利.结果血浆中阿克他利最低检测浓度0.098 mg·L-1,检测限4.9 ng,Acta在0.098~12.5 mg·L-1之间呈线性关系,回归方程y=32315.5X-100.9(r=0.9994,P<0.05),回收率在86.79%~90.7%,日间、日内变系数均在7.29%以下,符合生物样品分析要求.结论能够利用该方法进行阿克他利的人体药代动力学试验和相对生物利用度的研究.
AimTo set up the method of high performance liquid chromatography (HPLC) for detection of actarit in plasma. MethodsThe concentration of actarit in plasma was detected by high performance liquid chromatography and SPD-6AV detector.Results Under the condition of the test, actarit in plasma was well separated,and didn’t have the obvious interactive peak. The result of detection can represent concentration of primary drug. In linear range 0.098~12.5 mg·L~ -1 ,there was a regressive equation -Y-=32315.5-X--100.9(-r-=0.9994,-P-<0.05).The lowest concentration was 0.098 mg·L~ -1 ,and the lowest limit of detection was 4.9 ng. The return rate of the method was 86.7±3.36%~90.7±1.06%; intra-day, inter-day relative standard deviation(RSD) were under 7.2%.Conclusion The method of high performance liquid chromatography (HPLC) for detection of actarit in plasma can satisfy analytical requirement of biological preparation.
