摘要
目的建立HPLC法测定血浆中胺碘酮浓度的方法.方法采用反相高效液相色谱法测人体胺碘酮血药浓度.C18柱,流动相为乙腈-冰醋酸-三乙胺(940:60:0.0033,V/V).检测波长242 nm.结果在0.5~8.0 mg·L-1浓度范围内线性良好,最低检测限为0.3 mg·L-1,平均回收率为80.20%~96.03%.日内误差小于6%,日间误差小于10%.并用HPLC法测定了26例患者服用胺碘酮后的体内血药浓度.结论HPLC法操作简便、快速、准确,可用于胺碘酮的临床监测.
Aim To develop RP-HPLC method for the determination of Amiodarone in human serum.Methods Chromatographic separation has been achieved on C_ 18 column with Acetonitrile:Acetic Acid Glacial:Triethylamine(940∶60∶0.0033,V/V) as the mobile phase.Detection was set at UV 242 nm.Results The calibration curve was linear between 0.5 mg·L~ -1 and 8.0 mg·L~ -1 with a regression coefficient of 0.999 6. The lower limit of detection of Amiodarone in serum was 0.3 g·ml~ -1 .The method recovery rate was 80.20%~96.03%.Inter-day RSD of Amiodarone was less than 6% and intra-day RSD of Amiodarone was less than 10%.This assay procedure has been applied to monitoring serum levels of Amiodarone in 26 patients who received administration of drug.Conclusion The method was simple,rapid and accurate.It could be successfully used for TDM.
