^(99)Tc^m标记葡萄糖^(99)Tc^m-EC-DG在S180肉瘤荷瘤小鼠体内的分布研究

摘要 目的研究99TcmECDG在S180肉瘤荷瘤小鼠体内的分布规律,为其用于临床肿瘤显像作基础研究。方法将S180肉瘤荷瘤小鼠48只分成8组,实验前小鼠禁食8h以上。尾静脉注射99TcmECDG3.7MBq(100μCi)后5min、15min、30min、1h、2h、4h、8h和24h放血处死,取心、肝、脾、肺、脑、肾、肌肉、骨、小肠、胃、血液和肿瘤等组织或器官,称重并测量其放射性,计算每克组织百分注射剂量率(%ID/g)及肿瘤与血液、肌肉、肺、肝的放射性比值。同时,荷瘤小鼠5只,禁食,尾静脉注射99TcmECDG18.5MBq(500μCi)后相同时间进行显像。结果肿瘤组织与血液、肌肉、肺、肝的放射性比值随时间的延长逐渐上升,在4h时都超过1.0。显像结果与生物分布实验结果一致,随时间的延长瘤体周围组织放射性逐渐下降,而瘤体在4h时显影清晰。结论99TcmECDG在S180肉瘤荷瘤小鼠体内的生物学分布结果表明,其可用于肿瘤的葡萄糖代谢显像。 Objective To evaluate the biodistribution of techntium-99m labeled glucose ^(99)Tc^m-EC-DG in S180 Sarcoma tumor-bearing mice. Methods Fourty eight S180 sarcoma tumor-bearing mice were divided into 8 groups and fasted more than 8 hours before the injection of ^(99)Tc^m-EC-DG.Five minutes,15 minutes, 30 minutes, l hour, 2 hours,4 hours, 8 hours and 24 hours after the injection of 3.7 MBq ^(99)Tc^m-EC-DG,these mice were sacrificed by cutting the carotid.Selected tissues were excised and weighed, and the radioactivity was measured.The biodistrbution of radiotracer in each tissue was calculated as per percentage of the injected dose per gram of wet tissue weight (%ID/g).The ratio of tumor to blood, tumor to muscle, tumor to lung and tumor to liver were calculated. Meanwhile , 5 tumor-bearing mice were imaged after the injection of 18.5 MBq ^(99) Tc^m-EC-DG at the same time. Result The radioactivity ratio of tumor to blood, tumor to muscle, tumor to lung and tumor to liver increased after the injection, and all exceeded 1.0 at the 4th hour after injection. The imaging results were that same as that provided by biodistribution data, the radioactivity of tissues around tumor were decreased gradually after injection,and the tumor focus was clearly visualized at the 4th hour after injection.Conclusion ^(99)Tc^m-EC-DG can be used as an imaging agent of tumor glucose metabolism.
